圣多美主教座堂，正式名称为恩宠圣母主教座堂（Sé Catedral de Nossa Senhora da Graça），位于圣多美市中心人民广场，毗邻总统府。教堂始建于15世纪末，由阿尔瓦罗·德·卡明哈船长主持，1534年成为教区所在地。1576年至1578年，葡萄牙国王塞巴斯蒂昂时期重建；1784年因受损再次修复；1814年在居民倡议下重建；1956年改为新罗马式风格。这座教堂是宗教活动中心，也是葡萄牙殖民建筑与当地宗教传统的历史见证。
